    Rapid biodiversity assessment of arthropods for monitoring average local species richness and related ecosystem services

    Rapid biodiversity assessment (RBA) is proposed as an affordable indicator for monitoring local species richness of arthropods and sustainability of related ecosystem services. The indicator is based on strictly standardised sampling procedures and the identification of parataxonomic units (morphospecies) instead of species identification. The collection of arthropods was optimized with regard to trap types, time and length of collecting period, selection of four out of seven weekly samples, and choice of counted taxa and trophic guilds. By measuring arthropod activity, RBA is an indicator for functional diversity. Over a period of 8years, average yearly numbers of morphospecies were assessed in Switzerland in 15 agricultural habitats, 15 managed forests, and in 12 unmanaged habitats ranging from protected lowland wetlands to Alpine meadows. The yearly RBA-trend in unmanaged habitats is used for assessing the influence of climate and weather on biodiversity, and as a reference for measuring the relative influences of recent management changes in agriculture and forestry. The average number of morphospecies per sampling station per year depends on temperature, and was only marginally significantly increasing over time in agriculture, but not in forestry or unmanaged areas. Three RBA indices considered to be relevant for maintaining ecosystem services were calculated from the average number of morphospecies per location per year: (1) indicator for ecological resilience and sustainability (all morphospecies); (2) indicator for pollinator diversity (taxa with a majority of pollinators) and (3) indicator for biocontrol diversity (ratio between carnivore and herbivore guilds

    A primate virus generates transformed human cells by fusion

    A model that explains both the origin and sporadic nature of cancer argues that cancer cells are a chance result of events that cause genomic and epigenetic variability. The prevailing view is that these events are mutations that affect chromosome segregation or stability. However, genomic and epigenetic variability is also triggered by cell fusion, which is often caused by viruses. Yet, cells fused by viruses are considered harmless because they die. We provide evidence that a primate virus uses both viral and exosomal proteins involved in cell fusion to produce transformed proliferating human cells. Although normal cells indeed fail to proliferate after fusion, expression of an oncogene or a mutated tumor suppressor p53 in just one of the fusion partners is sufficient to produce heterogeneous progeny. We also show that this virus can produce viable oncogenically transformed cells by fusing cells that are otherwise destined to die. Therefore, we argue that viruses can contribute to carcinogenesis by fusing cells

    Response of arthropod species richness and functional groups to urban habitat structure and management

    Urban areas are a particular landscape matrix characterized by a fine-grained spatial arrangement of very diverse habitats (urban mosaic). We investigated arthropods to analyse biodiversity-habitat associations along five environmental gradients (age, impervious area, management, configuration, composition) in three Swiss cities (96 study sites). We considered total species richness and species richness within different functional groups (zoophagous, phytophagous, pollinator, low mobility, and high mobility species). Information theoretical model selection procedures were applied and predictions were calculated based on weighted models. Urban areas yielded on average 284 arthropod species (range: 169-361), with species richness correlating mostly with heterogeneity indices (configuration and composition). Species richness also increased with age of urban settlement, while enlarged proportions of impervious area and intensified habitat management was negatively correlated. Functional groups showed contrasted, specific responses to environmental variables. Overall, we found surprisingly little variation in species richness along the gradients, which is possibly due to the fine-grained spatial interlinkage of good (heterogeneous) and bad (sealed) habitats. The highly fragmented nature of urban areas may not represent a major obstacle for the arthropods currently existing in cities because they have probably been selected for tolerance to fragmentation and for high colonisation potential. Given that built areas are becoming denser, increasing spatial heterogeneity of the urban green offers potential for counteracting the detrimental effects of densification upon urban biodiversity.     Control of Pre-mRNA Splicing by the General Splicing Factors PUF60 and U2AF65

    Pre-mRNA splicing is a crucial step in gene expression, and accurate recognition of splice sites is an essential part of this process. Splice sites with weak matches to the consensus sequences are common, though it is not clear how such sites are efficiently utilized. Using an in vitro splicing-complementation approach, we identified PUF60 as a factor that promotes splicing of an intron with a weak 3' splice-site. PUF60 has homology to U2AF(65), a general splicing factor that facilitates 3' splice-site recognition at the early stages of spliceosome assembly. We demonstrate that PUF60 can functionally substitute for U2AF(65)in vitro, but splicing is strongly stimulated by the presence of both proteins. Reduction of either PUF60 or U2AF(65) in cells alters the splicing pattern of endogenous transcripts, consistent with the idea that regulation of PUF60 and U2AF(65) levels can dictate alternative splicing patterns. Our results indicate that recognition of 3' splice sites involves different U2AF-like molecules, and that modulation of these general splicing factors can have profound effects on splicing

    High-risk HPV E5-induced cell fusion: a critical initiating event in the early stage of HPV-associated cervical cancer

    <p>Abstract</p> <p>Background</p> <p>Cervical cancer is strongly associated with high-risk human papillomavirus (HPV) and viral oncoproteins E5, E6 and E7 can transform cells by various mechanisms. It is proposed that oncogenic virus-induced cell fusion may contribute to oncogenesis if p53 or apoptosis is perturbed simultaneously. Recently, HPV-16 E5 was found to be necessary and sufficient for the formation of tetraploid cells, which are frequently found in precancerous cervical lesions and its formation is strongly associated with HPV state.</p> <p>Presentation of the hypothesis</p> <p>We propose that high-risk HPV E5-induced cell fusion is a critical initiating event in the early stage of HPV-associated cervical cancer.</p> <p>Testing the hypothesis</p> <p>Our hypothesis can be tested by comparing the likelihood for colony formation or tumorigenic ability in nude mice between normal HaCaT cells expressing all three oncogenic proteins and E5-induced bi-nucleated HaCaT cells expressing E6 and E7. Moreover, investigating premature chromosome condensation (PCC) in HPV-positive and negative precancerous cervical cells is another way to assess this hypothesis.</p> <p>Implication of the hypothesis</p> <p>This viewpoint would change our understanding of the mechanisms by which HPV induces cervical cancer. According to this hypothesis, blocking E5-induced cell fusion is a promising way to prevent the progression of cervical cancer. Additionally, establishment of a role of cell fusion in cervical carcinogenesis is of reference value for understanding the pathogenesis of other virus-associated cancers.</p

    Tracking Ca2+ ATPase intermediates in real time by x-ray solution scattering

    Sarco/endoplasmic reticulum Ca2+ ATPase (SERCA) transporters regulate calcium signaling by active calcium ion reuptake to internal stores. Structural transitions associated with transport have been characterized by x-ray crystallography, but critical intermediates involved in the accessibility switch across the membrane are missing. We combined time-resolved x-ray solution scattering (TR-XSS) experiments and molecular dynamics (MD) simulations for real-time tracking of concerted SERCA reaction cycle dynamics in the native membrane. The equilibrium [Ca2] E1 state before laser activation differed in the domain arrangement compared with crystal structures, and following laser-induced release of caged ATP, a 1.5-ms intermediate was formed that showed closure of the cytoplasmic domains typical of E1 states with bound Ca2+ and ATP. A subsequent 13-ms transient state showed a previously unresolved actuator (A) domain arrangement that exposed the ADP-binding site after phosphorylation. Hence, the obtained TR-XSS models determine the relative timing of so-far elusive domain rearrangements in a native environment

    Biodiversity and structure of spider communities along a metal pollution gradient

    The objective of the study was to determine whether long-term metal pollution affects communities of epigeal spiders (Aranea), studied at three taxonomic levels: species, genera, and families. Biodiversity was defined by three indices: the Hierarchical Richness Index (HRI), Margalef index (DM) and Pielou evenness index (J). In different ways the indices describe taxa richness and the distribution of individuals among taxa. The dominance pattern of the communities was described with four measures: number of dominant species at a site, percentage of dominant species at a site, average dominant species abundance at a site, and the share of the most numerous species (Alopecosa cuneata) at a site. Spiders were collected along a metal pollution gradient in southern Poland, extending ca. 33 km from zinc and lead smelter to an uncontaminated area. The zinc concentration in soil was used as the pollution index.The study revealed a significant effect of metal pollution on spider biodiversity as described by HRI for species (p = 0.039), genera (p = 0.0041) and families (p = 0.0147), and by DM for genera (p = 0.0259) and families (p = 0.0028). HRI correlated negatively with pollution level, while DM correlated positively. This means that although broadly described HRI diversity decreased with increasing pollution level, species richness increased with increasing contamination. Mesophilic meadows were generally richer. Pielou (J) did not show any significant correlations. There were a few evidences for the intermediate disturbance hypothesis: certain indices reached their highest values at moderate pollution levels rather than at the cleanest or most polluted sites

    Species identification by experts and non-experts: comparing images from field guides

    Accurate species identification is fundamental when recording ecological data. However, the ability to correctly identify organisms visually is rarely questioned. We investigated how experts and non-experts compared in the identification of bumblebees, a group of insects of considerable conservation concern. Experts and non-experts were asked whether two concurrent bumblebee images depicted the same or two different species. Overall accuracy was below 60% and comparable for experts and non-experts. However, experts were more consistent in their answers when the same images were repeated, and more cautious in committing to a definitive answer. Our findings demonstrate the difficulty of correctly identifying bumblebees using images from field guides. Such error rates need to be accounted for when interpreting species data, whether or not they have been collected by experts. We suggest that investigation of how experts and non-experts make observations should be incorporated into study design, and could be used to improve training in species identification
